Overview

Clazakizumab is a high-affinity, humanized monoclonal antibody (IgG1) that targets and binds to the pro-inflammatory cytokine interleukin-6 (IL-6), thereby blocking its activity and preventing its interaction with the IL-6 receptor. This action inhibits both classical signaling via membrane-bound IL-6R and trans-signaling via soluble IL-6R. By reducing IL-6 activity, clazakizumab modulates immune responses and inflammation. It has been investigated primarily for chronic active antibody-mediated rejection (AMR) in kidney transplant recipients but has also been studied in conditions such as rheumatoid arthritis, psoriatic arthritis, COVID-19 infection (cytokine storm), severe asthma, anemia of chronic disease, cachexia, fatigue associated with cancer or chronic illness, Crohn's disease, graft-versus-host disease (GVHD), and general inflammation in older adults. Clazakizumab is notable for lacking both antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity and does not crosslink surface receptors[1][3][4][5][7][8].
