Clazakizumab + methotrexate is a combination therapy consisting of clazakizumab, a humanized monoclonal antibody that targets interleukin-6 (IL-6) cytokine, and methotrexate, a disease-modifying antirheumatic drug (DMARD). This combination has been studied for the treatment of rheumatoid arthritis (RA) in patients with an inadequate response to methotrexate alone. Clazakizumab binds to IL-6 with high affinity and blocks both classical signaling via membrane-bound IL-6R and trans-signaling via soluble IL-6R, while methotrexate further inhibits IL-6, providing complementary mechanisms of action. Clinical trials have shown that this combination significantly improves ACR20, ACR50, and ACR70 response rates compared to methotrexate monotherapy.
