Drug intelligence / Profile preview
CLB-001 is an engineered transfer RNA (tRNA) therapeutic containing 5-fluorouracil (5-FU) developed by Cloverleaf Bio. It is designed as a competitive substrate-mimic inhibitor that targets and irreversibly inhibits multiple tRNA-modifying enzymes, which cancer cells are abnormally dependent on to sustain high-throughput translation of oncogenic proteins. In preclinical studies, CLB-001 has demonstrated nanomolar potency and high selectivity for cancer cells over healthy cells, outperforming standard of care therapies in models of hepatocellular carcinoma (HCC) and lung adenocarcinoma without showing signs of systemic toxicity.
