Overview

CLBR001 is an autologous, engineered T cell product that forms the cellular component of a novel "switchable" chimeric antigen receptor T cell (sCAR-T) therapy platform. Unlike conventional CAR-T therapies, which are constitutively active, CLBR001 requires activation by a separately administered antibody-based adapter molecule (such as SWI019 or ABBV-461). This modular design allows for precise control over CAR-T activity and potentially improves safety and efficacy by enabling clinicians to modulate the therapy in real time. The primary mechanism involves targeting CD19-expressing cells when paired with an appropriate adapter molecule. Developed by Calibr-Skaggs Institute for Innovative Medicines (the drug development division of Scripps Research), in collaboration with AbbVie for some indications, CLBR001 is being investigated primarily for hematological malignancies such as B-cell malignancies and solid tumors like metastatic breast cancer. Early clinical data show promising response rates and improved safety profiles compared to traditional CAR-T therapies[1][5][6][7].
