Overview

CLD-101 (NSC-CRAd-S-pk7) is an investigational cell-based oncolytic virotherapy developed for the treatment of high-grade gliomas. It consists of neural stem cells (NSCs) that serve as a delivery vehicle for a conditionally replicating oncolytic adenovirus (CRAd). The adenovirus is engineered with a survivin promoter (S) to drive replication specifically in cancer cells and a pk7 fiber modification to enhance viral entry into tumor cells. The NSCs are progenitor cells capable of migrating through the brain parenchyma to reach invasive tumor cells, effectively bypassing the blood-brain barrier. Once at the tumor site, the NSCs release the oncolytic virus, which selectively infects and lyses glioma cells, potentially eliciting a systemic anti-tumor immune response. The therapy was originally developed at Northwestern University and is currently licensed to Calidi Biotherapeutics.
