Overview

CLD-423 is a first-in-class bispecific monoclonal antibody designed to simultaneously target the p19 subunit of interleukin-23 (IL-23p19) and tumor necrosis factor-like ligand 1A (TL1A). By inhibiting these two distinct pro-inflammatory pathways, CLD-423 aims to provide superior therapeutic efficacy in the treatment of inflammatory bowel disease (IBD) compared to single-target biologics. The asset was originally developed by Qyuns Therapeutics and was licensed to Caldera Therapeutics in April 2025 for exclusive global development and commercialization. The dual-mechanism approach is intended to suppress both the Th17-mediated immune response and fibro-inflammatory signaling, potentially addressing the complex pathology of Crohn's disease and ulcerative colitis.
