Drug intelligence / Profile preview
CLDN1-targeted antibody-drug conjugates (ADCs) represent an emerging class of targeted cancer therapeutics designed to selectively deliver cytotoxic payloads to tumor cells overexpressing Claudin-1 (CLDN1). Claudin-1 is a tight junction protein that is typically sequestered in normal epithelial tissues but becomes overexpressed and exposed on the cell surface in various solid tumors, including squamous non-small cell lung cancer (sqNSCLC), head and neck squamous cell carcinoma (HNSCC), cervical cancer, and esophageal cancer. By binding to exposed CLDN1, these ADCs are internalized, releasing their cytotoxic payloads (such as tubulin inhibitors or topoisomerase I inhibitors) to induce tumor cell death while sparing healthy tissues where CLDN1 is inaccessible. Prominent clinical-stage candidates in this class include Alentis Therapeutics' ALE.P02 (a tubulin inhibitor-based ADC) and ALE.P03 (a topoisomerase I inhibitor-based ADC).
