Drug intelligence / Profile preview
CLDN18.2-307-ADC (also known as TORL-2-307-ADC) is an investigational antibody-drug conjugate (ADC) targeting Claudin 18.2 (CLDN18.2), a tight junction protein that is aberrantly expressed in several solid tumors, particularly gastric, gastroesophageal junction, and pancreatic cancers. Developed by TORL BioTherapeutics in collaboration with the Slamon Research Lab at the University of California, Los Angeles (UCLA), the drug consists of a humanized monoclonal antibody (CLDN18.2-307-mAb) conjugated to the microtubule-disrupting cytotoxic payload monomethyl auristatin E (MMAE) via a protease-cleavable linker. Upon binding to CLDN18.2 on the surface of tumor cells, the ADC is internalized and trafficked to the lysosomal compartment, where MMAE is released to inhibit tubulin polymerization, leading to G2/M cell cycle arrest and apoptosis. CLDN18.2-307-ADC is currently being evaluated in Phase I clinical trials for patients with advanced CLDN18.2-positive solid tumors.
