The CLDN18.2 antibody is a monoclonal antibody developed by NovaRock Biotherapeutics, specifically engineered to target the first extracellular domain of Claudin 18.2 (CLDN18.2). CLDN18.2 is a four-transmembrane-domain tight junction protein that is selectively overexpressed in various solid tumors, particularly gastric and pancreatic cancers, while remaining largely absent from normal tissues. This antibody is characterized by its high affinity and efficient internalization upon binding to CLDN18.2-expressing cells, making it an ideal targeting component for antibody-drug conjugates (ADCs). NovaRock licensed this antibody to LigaChem Biosciences (formerly LegoChem Biosciences) for the development of next-generation ADCs, such as LCB02A, which utilize LigaChem's proprietary site-specific conjugation platform. The program is currently being evaluated in Phase 1/2 clinical trials for the treatment of CLDN18.2-positive advanced solid tumors.
