CLDN18.2-CD137 is a bispecific antibody (BsAb) being developed by NovaRock Biotherapeutics for the treatment of solid tumors, including gastric and pancreatic cancers. The molecule is designed to simultaneously bind to Claudin 18.2 (CLDN18.2), a protein highly expressed in several gastrointestinal cancers, and CD137 (4-1BB), a potent costimulatory receptor found on activated T cells and natural killer (NK) cells. By targeting both, the drug aims to concentrate CD137-mediated immune activation within the tumor microenvironment, thereby enhancing anti-tumor efficacy while reducing the systemic side effects typically associated with traditional CD137 agonists. The program is currently in the IND-enabling stage of development.
