CLDN18.2 SIR-T is an investigational cell therapy being developed by Angeles Therapeutics for the treatment of solid tumors expressing Claudin 18.2 (CLDN18.2). The therapy utilizes the proprietary Synthetic Immune Receptor T-cell (SIR-T) platform, which is designed to mimic the natural T-cell receptor (TCR) signaling complex. Unlike traditional Chimeric Antigen Receptor (CAR) T-cells that often rely on non-physiological signaling domains, SIR-T cells integrate synthetic receptors into the endogenous TCR complex, potentially offering superior sensitivity, persistence, and controlled activation within the immunosuppressive solid tumor microenvironment. CLDN18.2 is a tight junction protein that is highly overexpressed in several gastrointestinal malignancies, including gastric, gastroesophageal junction, and pancreatic adenocarcinomas, while maintaining restricted expression in healthy tissues. The program is currently in the IND-enabling phase of development.
