CLDN6-ADC is a preclinical-stage antibody-drug conjugate (ADC) being developed by NovaRock Biotherapeutics. It targets Claudin-6 (CLDN6), a tight junction protein that is highly expressed in several solid tumors—including ovarian, endometrial, testicular, lung, and gastric cancers—but has limited expression in normal adult tissues. The ADC consists of a CLDN6-specific monoclonal antibody conjugated to a cytotoxic payload. Upon binding to CLDN6 on the tumor cell surface, the complex is internalized, leading to the intracellular release of the payload and subsequent cell death. This mechanism is designed to provide potent anti-tumor activity while minimizing systemic toxicity.
