CLDN6 modRNA-LPX is an investigational mRNA-based cancer vaccine designed to encode the oncofetal antigen Claudin 6 (CLDN6), which is highly and specifically expressed in many solid tumors but not in healthy adult tissues. The drug consists of a modified RNA (modRNA) encoding CLDN6 formulated within lipoplexes (LPX) for delivery. Its primary mechanism involves promoting CLDN6 expression on dendritic cells after administration, thereby stimulating and enhancing the expansion and persistence of CLDN6-targeted chimeric antigen receptor T cells (CAR-T) in patients with relapsed or refractory advanced solid tumors. This approach aims to improve anti-tumor immune responses by combining CAR-T cell therapy with an mRNA vaccine that amplifies CAR-T cell activity. The drug is being developed primarily for use alongside CLDN6 CAR-T cell therapy in cancers such as germ cell tumors, epithelial ovarian cancer, endometrial carcinoma, and other solid tumor indications.
