CLDN6 SIR-T is an investigational cell therapy being developed by Angeles Therapeutics for the treatment of solid tumors expressing Claudin-6 (CLDN6). The therapy utilizes the company's proprietary Synthetic Immune Receptor T-cell (SIR-T) platform, which engineers T cells to express a synthetic receptor designed to mimic the natural T-cell receptor (TCR) complex. Unlike traditional Chimeric Antigen Receptor (CAR) T-cell therapies that often use single-chain variable fragments (scFv) and simplified signaling domains, the SIR-T architecture is intended to provide more physiological signaling, improved sensitivity to low-density antigens, and enhanced persistence within the tumor microenvironment. CLDN6 is a highly specific oncofetal antigen that is frequently overexpressed in various solid tumors, including ovarian, germ cell, and non-small cell lung cancers, but is virtually absent in healthy adult tissues, making it an ideal target for minimizing off-target toxicities. The program is currently in the IND-enabling phase of development.
