Overview

CLEC2A CAR T-cell therapy is an investigational second-generation chimeric antigen receptor (CAR) T-cell immunotherapy targeting the C-type lectin domain family 2 member A (CLEC2A) antigen. Developed by the Fred Hutchinson Cancer Center in collaboration with the University of Pittsburgh and Seattle Children's Hospital, this therapy is specifically designed for KMT2A-rearranged (KMT2A-r) acute myeloid leukemia (AML). CLEC2A is a tumor-specific antigen highly expressed in KMT2A-r AML but absent in normal hematopoietic cells and healthy tissues, minimizing on-target, off-tumor toxicity. The CAR construct utilizes a fully human binder (3H10), an IgG4 hinge, a 4-1BB costimulatory domain, and a CD3ζ signaling domain. Preclinical studies have demonstrated significant efficacy in patient-derived xenograft models, and clinical trials are anticipated to begin in early 2026.
