Immunitas Therapeutics' CLEC2D-TLR9 ISAC is an investigational immune-stimulating antibody complex (ISAC) designed for cancer immunotherapy, particularly for tumors with low immunological activity or poor T-cell infiltration. The molecule consists of a fully human monoclonal antibody targeting CLEC2D (also known as LLT1) conjugated to a CpG oligonucleotide payload, which acts as a Toll-like receptor 9 (TLR9) agonist. By targeting CLEC2D, which is expressed on tumor cells and immunosuppressive myeloid cells, the complex delivers the TLR9 agonist specifically to the tumor microenvironment. This targeted delivery triggers the activation of myeloid cells, B cells, and plasmacytoid dendritic cells, promoting an inflammatory response that enhances the recruitment and function of T cells and NK cells, potentially turning "cold" tumors into immunologically "hot" ones.
