Overview

CLEC5A siRNA is a research-grade small interfering RNA (siRNA) designed to silence the expression of the C-type lectin domain family 5 member A (CLEC5A) gene, also known as Myeloid DAP12-associating lectin 1 (MDL-1). By triggering the degradation of CLEC5A mRNA, this RNA interference (RNAi) agent knocks down the CLEC5A receptor on myeloid cells, particularly macrophages. CLEC5A is a critical mediator of the inflammatory response; its activation by viruses like dengue and Japanese encephalitis virus (JEV) triggers proinflammatory cytokine release. In oncology, CLEC5A has been identified as a regulator of tumor-associated macrophage (TAM) polarization and immunosuppression in the glioblastoma (GBM) microenvironment. While explored in preclinical models for viral infections and glioblastoma, it currently exists primarily as a research tool rather than a clinical-stage therapeutic.
