Overview

Cleistanthin A is a naturally occurring arylnaphthalide lignan glycoside isolated primarily from the plant Cleistanthus collinus. It is one of the toxic principles of this plant and has been studied for its pharmacological activities including antihypertensive effects (alpha adrenergic antagonism), diuretic action, and potential anticancer properties. Mechanistically, cleistanthin A acts as a noncompetitive antagonist at alpha‑adrenergic receptors (notably α₁), leading to vasodilatory effects. It also exhibits anti-inflammatory and antioxidant activities and has shown efficacy in preclinical models for reducing benign prostatic hyperplasia by downregulating inflammatory pathways (such as IL‑6/STAT‑3/cyclin D1) and inducing apoptosis in target tissues[1][4][7][10].
