Overview

Clenoliximab is a chimeric monoclonal antibody (IgG4 isotype) derived from *Macaca irus* and *Homo sapiens* that targets the CD4 molecule on the surface of T lymphocytes. It acts as an immunomodulator by binding to CD4, leading to down-modulation of CD4 density on T cells without depleting them. This mechanism involves antibody-mediated stripping of CD4 from the cell surface, which inhibits antigen-induced T-cell proliferation, lymphokine release, and helper T-cell functions. Clenoliximab was investigated primarily for the treatment of rheumatoid arthritis, where T cell activation drives inflammation and tissue damage. The drug was developed as a derivative of keliximab with reduced Fc receptor affinity due to its IgG4 backbone and specific amino acid substitutions[1][2][4][6].
