Overview

**CLG561 + LFG316** is an investigational combination therapy for geographic atrophy (GA) secondary to age-related macular degeneration (AMD), administered as repeated intravitreal injections. CLG561 is a monoclonal antibody targeting properdin, a positive regulator of the alternative complement pathway, while LFG316 (also known as tesidolumab) is a monoclonal antibody inhibitor of complement factor C5. Both drugs seek to modulate complement activity in the retina to slow progression of GA. The combination was developed to explore synergistic inhibition of different steps in the complement cascade. While preclinical and early clinical studies indicated safety for both drugs, a phase 2 clinical trial failed to show significant efficacy over sham in reducing GA lesion growth.
