Overview

Clinafloxacin is a synthetic fluoroquinolone antibiotic with broad-spectrum activity against Gram-positive, Gram-negative, and anaerobic bacteria. Its mechanism of action involves inhibition of bacterial type II topoisomerases—DNA gyrase and topoisomerase IV—enzymes essential for DNA replication and transcription. By stabilizing the enzyme-DNA cleavage complexes, clinafloxacin induces double-stranded DNA breaks that ultimately lead to bacterial cell death[3][6][8]. Clinafloxacin demonstrated higher activity against certain pathogens (including *Streptococcus pneumoniae* and *Pseudomonas aeruginosa*) compared to other fluoroquinolones[3][6]. Despite promising efficacy in clinical trials for serious infections such as sepsis, its development was discontinued due to safety concerns including hypoglycemia (dangerously low blood sugar), photosensitivity reactions (light sensitivity), and significant drug-drug interactions[1][3]. The drug was originally developed in the 1990s but never reached market approval.
