clindamycin phosphate + adapalene

IDP-126 Component C is a topical combination gel consisting of clindamycin phosphate (1.2%) and adapalene (0.15%). Developed by Bausch Health (through its Ortho Dermatologics division), it served as one of the active dyad comparator arms in the Phase 2 clinical evaluation of IDP-126 (now marketed as Cabtreo), which is a triple-combination gel for the treatment of acne vulgaris. Clindamycin phosphate is a lincosamide antibiotic that inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit of *Cutibacterium acnes*. Adapalene is a third-generation retinoid that modulates cellular differentiation, keratinization, and inflammatory processes by acting as a selective agonist for retinoic acid receptors (RAR-beta and RAR-gamma). This specific combination was used to demonstrate the incremental efficacy of adding benzoyl peroxide to create the final triple-combination product.
