CLIO-ICT is an investigational **MMP-14-activated theranostic nanoparticle** developed for cancer treatment and magnetic-resonance imaging monitoring. It comprises a cross-linked iron oxide nanoparticle core based on ferumoxytol, an MMP-14-cleavable peptide linker, and the vascular-disrupting azademethylcolchicine payload ICT3105. In MMP-14-expressing tumors, linker cleavage releases the tubulin-targeting payload, disrupting tumor endothelium and vasculature and promoting apoptosis of glioblastoma-initiating cells; the iron oxide core enables MRI tracking of tumor delivery.
