Overview

Clitocybe nebularis lectin (CNL) is a fungal protein isolated from the edible mushroom *Clitocybe nebularis* (clouded agaric). It functions as a glycan-binding protein with high specificity for N-acetylgalactosamine (GalNAc) motifs, particularly the Tn antigen (GalNAc-α-Ser/Thr) and the sialyl-Tn (STn) antigen. These truncated O-glycans are tumor-associated carbohydrate antigens (TACAs) that are frequently overexpressed in various cancers due to aberrant glycosylation processes. Research conducted by institutions such as the University of Tsukuba and the National Institute of Advanced Industrial Science and Technology (AIST) has highlighted CNL's ability to selectively stain pancreatic cancer cells while showing minimal reactivity with normal pancreatic ductal epithelium. This specificity makes CNL a promising candidate for the development of diagnostic biomarkers and targeted therapeutic strategies for pancreatic and other solid tumors.
