Overview

Clivatuzumab (PAM4) is a humanized monoclonal antibody that targets a specific epitope of Mucin-1 (MUC1), a glycoprotein that is highly expressed in pancreatic ductal adenocarcinoma but not in normal pancreatic tissue or pancreatitis. Developed by Immunomedics, the antibody was primarily developed as a radiopharmaceutical conjugate, clivatuzumab tetraxetan (IMMU-107), which is labeled with Yttrium-90 (90Y) for the treatment of advanced pancreatic cancer. Although it showed promise in early-phase trials and reached Phase 3 clinical development (the PANCRIT-1 trial), the program was discontinued after the trial failed to demonstrate a significant improvement in overall survival.
