Anti-CLL-1 CAR NK cells are natural killer (NK) cells that have been genetically engineered to express a chimeric antigen receptor (CAR) targeting C-type lectin-like molecule 1 (CLL1), an antigen highly expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells but not on healthy hematopoietic stem cells. The addition of the anti-CLL-1 CAR enables these NK cells to specifically recognize and kill AML cancerous cells with improved specificity. Compared to traditional T cell-based therapies, such as CAR-T, anti-CLL-1 CAR NK cell therapy offers advantages including a lower risk of cytokine release syndrome, neurotoxicity, and graft-versus-host disease. These therapies are being developed as "off-the-shelf" allogeneic products for the treatment of AML[3][4][1].
