Overview

CLL-1 nanobody CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target C-type lectin-like molecule-1 (CLL-1), also known as CLEC12A. Developed by researchers at the Sino-American Cancer Foundation and collaborating institutions, this biologic utilizes llama-derived single-domain antibodies (nanobodies or VHH) as the antigen-binding domain instead of the traditional single-chain variable fragments (scFv). CLL-1 is a type II transmembrane glycoprotein highly expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells but minimally expressed on healthy hematopoietic stem cells. By leveraging the high specificity and low immunogenicity of nanobodies, this therapy aims to provide potent anti-leukemic activity while reducing on-target, off-tumor toxicities, particularly the depletion of normal hematopoietic progenitors. Preclinical studies have demonstrated rapid and durable cytotoxicity in AML models and the preservation of normal colony-forming capacity.
