CLL1 CAR-γδ T is an investigational allogeneic chimeric antigen receptor (CAR) cell therapy that utilizes gamma delta (γδ) T cells engineered to target C-type lectin-like molecule-1 (CLL-1, also known as CLEC12A). CLL-1 is a cell surface glycoprotein frequently overexpressed on acute myeloid leukemia (AML) blasts and leukemic stem cells (LSCs), while remaining largely absent on normal hematopoietic stem cells, making it a selective target for myeloid malignancies. By employing γδ T cells instead of conventional alpha beta (αβ) T cells, this therapy leverages the innate anti-tumor activity of γδ T cells and their MHC-independent recognition, which significantly reduces the risk of graft-versus-host disease (GvHD) and supports an "off-the-shelf" allogeneic manufacturing model. In preclinical research, CLL1 CAR-γδ T cells have demonstrated potent cytotoxicity against CLL-1-positive AML cell lines and are used as a benchmark for evaluating next-generation γδ T cell platforms.
