CLL1 CAR-DNT is an allogeneic, "off-the-shelf" cell therapy consisting of double negative T (DNT) cells engineered to express a chimeric antigen receptor (CAR) targeting C-type lectin-like molecule-1 (CLL1; CLEC12A). Developed by Juventas Cell Therapy, the product is intended for the treatment of relapsed or refractory acute myeloid leukemia (AML). DNT cells, defined as CD3+CD4-CD8- T cells, are utilized for their innate ability to kill AML cells and their low propensity for inducing graft-versus-host disease (GvHD) in an allogeneic setting. The CAR construct incorporates a humanized scFv (H1-3Q2) and signaling domains such as 4-1BB and CD3ζ. Preclinical studies have demonstrated that these cells can effectively lyse CLL1-expressing AML cells and leukemia stem cells while sparing hematopoietic stem cells.
