CLL1 CAR-NK cells are a type of chimeric antigen receptor (CAR) natural killer (NK) cell therapy under development for the treatment of acute myeloid leukemia (AML). These NK cells are genetically modified to express a CAR specifically targeting CLL-1 (encoded by CLEC12A), an antigen highly expressed on malignant AML cells but absent on normal hematopoietic stem and progenitor cells. The therapy aims to leverage the inherent anti-tumor properties of NK cells while enhancing their specificity and potency against AML. Different CAR designs with varying spacer structures have been investigated, with structurally optimized versions, particularly those incorporating autocrine cytokine stimulation (e.g., IL-2 or IL-15), showing enhanced cytotoxicity, persistence, and anti-leukemic efficacy in preclinical models.
