This combination cell therapy consists of two distinct chimeric antigen receptor (CAR) T-cell products: one targeting C-type lectin-like molecule-1 (CLL1) and another dual-targeting CD33 and CD123. Developed by The First Affiliated Hospital of Zhejiang University, this cocktail or sequential infusion approach is designed to overcome antigen escape and tumor heterogeneity in patients with relapsed or refractory acute myeloid leukemia (AML). CLL1, CD33, and CD123 are highly expressed on AML blasts and leukemic stem cells (LSCs), but their expression varies among patients and clones. By targeting three distinct antigens, the therapy aims to achieve deeper remissions and reduce the likelihood of relapse compared to single-target CAR-T therapies. The treatment typically involves the sequential administration of the two CAR-T products following lymphodepleting chemotherapy.
