Drug intelligence / Profile preview
CLL1-CD33 compound CAR (cCAR) is an investigational dual-targeting chimeric antigen receptor (CAR) T-cell therapy being developed for the treatment of acute myeloid leukemia (AML). This biologic therapy consists of two complete CAR constructs connected by a self-cleaving P2A peptide, allowing for the simultaneous targeting of two distinct antigens: CD33, which is expressed on bulk AML disease cells, and CLL-1 (C-type lectin-like molecule-1), which is associated with leukemic stem cells and disease relapse. The therapy is designed to address intra-tumor heterogeneity and mitigate the risk of antigen escape. Additionally, the construct incorporates a CD52 safety switch, which enables the rapid termination of the CAR-T cells using the monoclonal antibody alemtuzumab if severe adverse events occur. Clinical trials, primarily conducted in China, have evaluated both autologous and HLA-matched sibling donor-derived versions of the therapy, showing promise in achieving complete remission with minimal residual disease negativity in patients with relapsed or refractory AML.
