Overview

CLL1-CD33 compound CAR T cells are a dual-targeting chimeric antigen receptor T cell therapy, engineered to recognize both CLL1 (CLEC12A) and CD33 antigens commonly expressed on acute myeloid leukemia (AML) cells and leukemia stem cells. These therapeutic T cells are typically generated from a patient's own (autologous) cells and are genetically modified to express two CAR constructs: one for CLL1 and one for CD33, linked by a self-cleaving peptide. This dual-targeted approach aims to increase anti-leukemic activity, reducing tumor escape seen with single-target CAR T therapies, and eradicate both leukemia blasts and their stem cells. Preclinical and early phase clinical trials in relapsed/refractory AML have shown promising results, including minimal residual disease (MRD) negativity and complete remissions. The therapy includes a safety switch for rapid CAR T cell elimination post-treatment. Potential indications may include other diseases with expression of CLL1 and CD33, like blast crisis in myelodysplastic syndrome or chronic myeloid leukemia[1][2][5][6].
