Drug intelligence / Profile preview
CLN-2501 is a first-in-class, selective smooth muscle myosin inhibitor being developed as a capillary-selective vasodilator to treat capillary no-reflow (CNR) following acute myocardial infarction (AMI). Developed by Motorpharma (a spinout of Eötvös Loránd University) and licensed to PeriFlow Therapeutics, CLN-2501 directly binds to smooth muscle myosin (specifically Myosin-11) in capillary pericytes, relaxing the pathological hypercontraction that restricts microvascular blood flow after reperfusion. In preclinical models of cardiac and renal ischemia, CLN-2501 rapidly reopened capillaries, restored blood flow, reduced infarct size, and improved survival without affecting systemic blood pressure. The drug is also being explored for other ischemic conditions, including acute kidney injury, ischemic stroke, and organ transplantation, and is advancing toward clinical development in the United States.
