Overview

CLN-418 is a clinical-stage bispecific monoclonal antibody that targets both B7H4 (a tumor-associated antigen highly expressed in multiple solid tumors) and 4-1BB (Tumor necrosis factor receptor superfamily member 9, a costimulatory molecule important for T and NK cell activation). The drug is designed to conditionally activate 4-1BB signaling only in the presence of B7H4-expressing tumor cells, thereby promoting targeted T cell activation and anti-tumor immune responses while minimizing off-target toxicity. This mechanism aims to improve safety compared to non-selective 4-1BB agonists. CLN-418 is being developed by Cullinan Oncology, with rights licensed from Harbour BioMed, and is currently being evaluated in a Phase 1 clinical trial for patients with advanced solid tumors[1][3][5][6].
