Overview

CLN-617 is a first-in-class, single-chain fusion protein designed for intratumoral injection in the treatment of advanced solid tumors. It uniquely combines two potent and synergistic antitumor cytokines—interleukin 2 (IL-2) and interleukin 12 (IL-12)—in a single molecule, along with leukocyte-associated immunoglobulin-like receptor 2 (LAIR2) and human serum albumin (HSA). LAIR2 binds to collagen in the tumor microenvironment, while HSA increases molecular weight; both features are intended to retain CLN-617 within the injected tumor, enhancing local efficacy and reducing systemic toxicity. The mechanism of action involves stimulating T cells and natural killer (NK) cells via IL-2 and IL-12 signaling pathways, leading to robust local and systemic antitumor immune responses. Preclinical studies demonstrated that intratumoral administration of CLN-617 eradicated both treated and untreated tumors in animal models, improved response to anti-PD1 checkpoint therapy, induced abscopal effects dependent on cellular immunity, and generated memory T cell responses. CLN-617 is currently being evaluated in Phase 1 clinical trials as monotherapy or combined with pembrolizumab for patients with advanced solid tumors[1][4][5][8].
