CLN-619 is a humanized IgG1 monoclonal antibody developed for the treatment of various cancers, including solid tumors and hematologic malignancies such as relapsed/refractory multiple myeloma. It targets the stress-induced ligands MHC class I polypeptide-related sequence A (MICA) and MHC class I polypeptide-related sequence B (MICB), which are often shed from tumor cells to evade immune detection. By binding to MICA/MICB, CLN-619 prevents their proteolytic cleavage and restores their expression on the tumor cell surface. This enhances recognition by NKG2D-expressing innate and adaptive immune cells, promoting antitumor activity through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). CLN-619 has demonstrated favorable safety profiles both as monotherapy and in combination with pembrolizumab in early-phase clinical trials, with objective responses observed across multiple tumor types[1][2][3][5][7].
