Clobetasol + spironolactone is a topical combination medication that has been studied for preventing skin atrophy associated with corticosteroid use. Clobetasol is a potent topical glucocorticoid that can cause skin thinning (epidermal atrophy) as a side effect. Spironolactone, a mineralocorticoid receptor antagonist, has been shown to limit this clobetasol-induced skin atrophy when co-applied. The combination works by allowing the therapeutic benefits of clobetasol while the spironolactone component helps protect against the skin-thinning side effects through mineralocorticoid receptor blockade.
