Overview

Clobetasone butyrate + tacrolimus is a topical combination therapy used primarily in dermatology for the treatment of atopic dermatitis and other inflammatory skin conditions. Clobetasone butyrate is a synthetic glucocorticoid corticosteroid that reduces inflammation, itching, and erythema by modulating gene expression through the glucocorticoid receptor. Tacrolimus is a macrolide immunosuppressant that acts as a calcineurin inhibitor, suppressing T-cell activation and cytokine release by inhibiting calcineurin via binding to FKBP-12. The combination aims to provide enhanced efficacy while minimizing steroid exposure, leveraging the anti-inflammatory effects of clobetasone with the steroid-sparing immunomodulatory action of tacrolimus. Clinical studies have shown this regimen can be more effective than either agent alone in improving symptoms of moderate to severe atopic dermatitis[1][6].
