Clocortolone caproate is a synthetic corticosteroid ester used primarily for its anti-inflammatory and antipruritic properties in dermatological conditions. It acts as a prodrug of clocortolone and exerts its effects by binding to the glucocorticoid receptor (NR3C1), leading to the induction of lipocortins that inhibit phospholipase A2. This inhibition reduces the release of arachidonic acid from membrane phospholipids and decreases synthesis of pro-inflammatory mediators such as prostaglandins and leukotrienes. The result is reduced inflammation and immune response in affected tissues. Clocortolone caproate is characterized by moderate potency as a topical corticosteroid with high lipid solubility for effective skin penetration[2][3][5]. Its primary indication is for corticosteroid-responsive dermatoses.
