Overview

A combination chemotherapy regimen consisting of clofarabine and cyclophosphamide. This combination demonstrates synergistic antileukemic effects, primarily used for treating relapsed or refractory acute leukemias in pediatric patients. Clofarabine inhibits DNA polymerase \u03b1 and ribonucleotide reductase, disrupts mitochondrial membrane integrity leading to programmed cell death, and inhibits DNA repair. Cyclophosphamide is an alkylating agent that cross-links DNA via active metabolites. The combination shows enhanced efficacy because clofarabine potentiates cyclophosphamide's activity by inhibiting the repair of cyclophosphamide-induced DNA damage.
