Overview

Clofibric acid (chlorophenoxyisobutyrate) is a monocarboxylic acid and the active metabolite of several lipid-lowering drugs in the fibrate class, including clofibrate. It is structurally defined as isobutyric acid substituted at position 2 by a p-chlorophenoxy group. Clofibric acid acts primarily as an agonist of peroxisome proliferator-activated receptor alpha (PPARα), leading to increased lipolysis and elimination of triglyceride-rich particles from plasma by activating lipoprotein lipase and reducing apolipoprotein C-III production. This results in decreased plasma cholesterol and triglyceride levels. The compound has been used clinically for hyperlipidemia but is now largely obsolete due to safety concerns[3][4][5][6].
