Overview

Clone2HM CAR-T is a second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target CD33, a transmembrane protein frequently overexpressed in acute myeloid leukemia (AML). Developed by researchers at Fujian Medical University, this therapy utilizes a humanized monoclonal antibody fragment (Clone2HM) as the antigen-binding domain, coupled with 4-1BB (CD137) costimulatory and CD3ζ signaling domains. In preclinical studies, Clone2HM CAR-T cells demonstrated potent, antigen-specific cytotoxicity and interferon-gamma (IFN-γ) production against CD33-positive AML cells. In vivo experiments using MOLM13-Luciferase xenograft models showed that Clone2HM CAR-T cells effectively suppressed tumor growth and provided durable disease control, outperforming benchmarks derived from gemtuzumab.
