Clone3HM-MMAE is an investigational **CD33-targeting antibody-drug conjugate** developed from a fully humanized anti-CD33 monoclonal-antibody clone, Clone3HM. The antibody is conjugated through a cleavable Mal-PEG8-VC-PAB linker to the antimitotic payload monomethyl auristatin E. It is intended for treatment of CD33-positive acute myeloid leukemia, where CD33-mediated binding and internalization are intended to release MMAE intracellularly, disrupt microtubules, and induce leukemic-cell death. In MOLM13-Luc acute myeloid leukemia xenograft models, a single 5 mg/kg intravenous dose produced marked tumor suppression and prolonged survival relative to gemtuzumab-MMAE without overt body-weight loss. The candidate remains at the preclinical stage and has no assigned INN or marketed brand name.
