Overview

Clorgiline is a small molecule, irreversible and selective inhibitor of monoamine oxidase A (MAO-A), structurally related to pargyline. It was developed as an antidepressant but was never marketed for clinical use. Clorgiline acts by inhibiting the enzyme MAO-A, which deaminates neurotransmitters such as serotonin and norepinephrine, thereby increasing their levels in the brain. In addition to its primary action on MAO-A, it binds with high affinity to the sigma-1 receptor and very high affinity to the imidazoline I2 receptor. Clorgiline has also been identified as a multidrug efflux pump inhibitor in research settings[1][2][3][4][5].
