Overview

**CLP-1001** is a non-absorbed oral polymer developed by Sorbent Therapeutics as a cross-linked polyelectrolyte (CLP) to treat fluid overload in heart failure patients. It binds excess sodium and fluid in the gastrointestinal tract, removing them independently of kidney function, thereby alleviating congestion symptoms without relying on diuretics. Positive results from a Phase 2a trial in 111 heart failure patients with chronic kidney disease showed improvements in signs and symptoms of fluid retention, prompting initiation of the Phase 2b STEPWISE trial in 2013, which randomized 250 patients on optimized heart failure therapy to 15g daily CLP-1001 or placebo, evaluating efficacy via metrics like 6-Minute Walk Test, body weight, Kansas City Cardiomyopathy Questionnaire, NT-proBNP, and time to hospitalization or decongestion.[1][2][3][4][6][10]
