It is a phospholipid drug conjugate (PDC) that utilizes iodine-125 as an Auger electron-emitting radioisotope, designed for targeted radiotherapy of solid tumors. The compound leverages Cellectar’s proprietary PDC platform to deliver the radioisotope directly into cancer cells, maximizing intracellular delivery and minimizing off-target effects. Auger emitters like iodine-125 are characterized by their extremely short emission range (a few nanometers), which allows for highly precise tumor cell killing with minimal damage to surrounding healthy tissue. Preclinical studies have demonstrated robust in vivo activity and tolerability in multiple animal models, including triple-negative breast cancer, lung cancer, and colorectal cancer. CLR 121125 has received IND clearance from the FDA and is set to enter Phase 1b/2a clinical trials in patients with triple-negative breast cancer[1][2][4][7].
