Overview

CLR 12120 is a preclinical, targeted alpha-emitting radioconjugate drug candidate developed by Cellectar Biosciences for the treatment of solid tumors and other neoplasms. It is part of a series of phospholipid ether (PLE)-based conjugates designed to deliver potent alpha-emitting radioisotopes—such as actinium-225, lead-212, or astatine-211—directly to malignant cells while minimizing off-target effects. The PLE platform targets unique microdomains in cancer cell membranes that are distinct from those in normal cells due to altered metabolic needs and membrane composition. Preclinical studies have demonstrated high tumor uptake, low off-target accumulation, and therapeutic efficacy in xenograft models of pancreatic and triple-negative breast cancer. The program aims to improve the therapeutic index by increasing delivery to tumor cells (including cancer stem cells) while reducing exposure to healthy tissues[1][2][4].
