Overview

CLR1401 is a synthetic alkylphosphocholine small-molecule designed as a cancer-targeting platform that selectively accumulates in tumor cell membranes and lipid rafts while sparing most normal tissues. It serves as the non‑radioactive phospholipid ether scaffold for a family of tumor‑targeting agents, including the radioiodinated derivatives 124I‑CLR1404 and 131I‑CLR1404 used for PET/SPECT imaging and targeted radiotherapy, respectively, which show prolonged and preferential retention in a wide range of solid tumors and cancer stem cell models in rodents and humans.[2][3][6] CLR1401 itself has been characterized as an anticancer agent in preclinical toxicokinetic and pharmacology studies and as a drug‑vehicle for conjugation with cytotoxic payloads (for example, radionuclides or small‑molecule drugs) to enable broad‑spectrum, tumor‑selective imaging and treatment of cancers.[2][3][9][10]
