Overview

CLR151 is a small molecule drug developed by Clera Inc. as a fast-off dopamine D2 receptor antagonist, intended primarily for central nervous system diseases such as schizophrenia. Preclinical studies in parkinsonian macaques demonstrated that CLR151 can significantly reduce L-DOPA-induced dyskinesia and psychosis-like behavior by antagonizing dopamine D2 receptors, with a rapid dissociation rate from the receptor. Despite significant reduction in dyskinesia and psychosis-like symptoms, CLR151 also decreased the duration of L-DOPA’s anti-parkinsonian effects and increased parkinsonian disability, indicating potential limitations in its therapeutic window. The drug has not advanced beyond preclinical development as of August 2025[5].
